Phenylethylmethylhydantoin Farmacologia
Mefenitoína es un fármaco antiepiléptico que puede ser útil en el tratamiento de la epilepsia. El principal lugar de acción parece ser la corteza motora, donde propagación de la actividad convulsiva es inhibida. Probablemente mediante la promoción de eflujo de sodio de las neuronas, mephenytoin tiende a estabilizar el umbral contra la hiperexcitabilidad causada por estimulación excesiva o cambios ambientales capaces de reducir el sodio de membrana gradiente. Esto incluye la reducción de la potenciación postetánica en las sinapsis. La pérdida de la potenciación postetánica evita los focos corticales de convulsiones detonar adyacentes áreas corticales. Mefenitoína reduce la máxima actividad de los centros del tallo cerebral responsables de la fase tónica de las crisis tónico-clónicas (gran mal), convulsiones.
