    Doxorubicin HCl Pharmacologie

    La doxorubicine est un antinéoplasique de la classe des anthracyclines. Propriétés générales des médicaments de cette classe comprennent: l'interaction avec l'ADN dans une variété de différentes façons, y compris d'intercalation (exprimage entre les paires de bases), la rupture des brins d'ADN et l'inhibition de l'enzyme topoisomérase II. La plupart de ces composés ont été isolés à partir de sources naturelles et les antibiotiques. Cependant, ils n'ont pas la spécificité des antibiotiques antimicrobiens et produire ainsi des effets toxiques importants. Les anthracyclines sont parmi les médicaments antitumoraux plus importants disponibles. La doxorubicine est largement utilisé pour le traitement de plusieurs tumeurs solides tout en daunorubicine et idarubicine sont utilisés exclusivement pour le traitement de la leucémie. La doxorubicine peut également inhiber l'activité de la polymérase, affectent régulation de l'expression des gènes, et de produire gratuitement les dommages des radicaux à l'ADN. Doxorubicine possède un effet antitumoral contre un large spectre de tumeurs, soit greffé ou spontané. Les anthracyclines sont pas spécifiques du cycle cellulaire.

    Patients and their caregivers should be informed of the expected adverse effects of DOXIL® , particularly hand-foot syndrome, stomatitis, and neutropenia and related complications of neutropenic fever, infection, and sepsis.

    Hand-Foot Syndrome (HFS): Patients who experience tingling or burning, redness, flaking, bothersome swelling, small blisters, or small sores on the palms of their hands or soles of their feet (symptoms of Hand-Foot Syndrome) should notify their physician.

    Stomatitis: Patients who experience painful redness, swelling, or sores in the mouth (symptoms of stomatitis) should notify their physician.

    Fever and Neutropenia Patients who develop a fever of 100.5°F or higher should notify their physician.

    Nausea, vomiting, tiredness, weakness, rash, or mild hair loss Patients who develop any of these symptoms should notify their physician.

    Following its administration, DOXIL® may impart a reddish orange color to the urine and other body fluids. This nontoxic reaction is due to the color of the product and will dissipate as the drug is eliminated from the body.
