Pharmacologie
Isradipine, le plus puissant des canaux calciques agent bloquant de la classe des dihydropyridines, est similaire à la nifédipine, l'amlodipine et la félodipine. Il se lie aux canaux calciques avec une haute affinité et spécificité et inhibe le flux de calcium dans le muscle cardiaque et lisse. Les effets observés dans des expériences in vitro et mécanistiques étudiées chez les animaux intacts et l'homme sont compatibles avec ce mécanisme d'action et sont typiques de la classe.

Absorption
Isradipine est de 90% -95% absorbée et est soumise à un effet de premier passage hépatique, résultant en une biodisponibilité d'environ 15% -24%.

Toxicite
Les symptômes de surdose comprennent la léthargie, la tachycardie sinusale et une hypotension transitoire. Létalité importante a été observée chez les souris ayant reçu des doses orales de plus de 200 mg / kg et des lapins ayant reçu environ 50 mg / kg de l'isradipine. Les rats ont toléré des doses de plus de 2000 mg / kg sans effet sur la survie.

Proper use of this medicine

Take this medicine exactly as directed. This medicine is normally taken once daily. It is
taken orally and may be taken with or without food. The extended-release tablet and capsule
should be swallowed whole. Do not chew, divide, or crush the extended-release tablet or
capsule.
Missed Dose

Take your next dose as soon as you remember. If it is time for your next dose, skip the
missed dose and go back to your regular schedule. Do not double doses.
Storage

Keep this medication in the container it came in, tightly closed, and out of reach of
children. Store it at room temperature and away from excess heat and moisture. Throw away
any medication that is outdated or no longer needed.

Pregnancy/Nursing

but animal studies suggest caution during pregnancy, and use should only be when risks are
outweighed by possible benefits. For nursing mothers, it is not known if amlodipine is
